If you had the option to choose your inital 4 card hand, what would you take? Obviously there isn't a "one size fits all", as different villains call for different tools, so feel free to be creative with it. 

I'll start with what one of the easier characters for such a list.

Waith: 2 Impromptu Inventions, Grappling Hook, MicroTargeting Computer: This pretty much gives you a complete first turn damage setup with a Grappling Hook as safety net. I'd take this hand vs any boss, any day.

Mr. Fixer: Driving Mantis, Dual Crowbars, Overdrive, Toolbox.

Slightly on the offense side, being able to deal either 1 target 4 damage or 2 targets 2 damage. Yets still able to toss some environment/villain damage on occasion for some protection. Early overdrive to ensure that Salvage Yard will be able to retrieve one. Toolbox to give the extra cards to get Charge/Overdrive/Bloody Knuckles or replacement tools or styles if needed.

Absolute Zero: Onboard Module Installation, Onboard Module Installation, Onboard Module Installation, Impale.

This gets both modules and some other useful stuff into your hand in addition to thinning out your deck, all on the first turn.  And who doesn't love a first-turn Impale?*

Ra: Summon Staff x4

Nightmist: Call Forth, Mists of Time 4, Master of Magic, literally anything else

Argent Adept: Alacritous Subdominant, Silver Shadow, Arcane Cadence (with the intent of getting an instrument) and Inventive Preparation.

Chrono-Ranger: Displaced Armory (hat), Sudden Contract, Sudden Contract, Displaced Armory

Some Targets: Assault Rifle, Hollow Points, Pride, Flak

Many Targets (Handle it): SMG, Shock Rounds, Hair Trigger, Assault Rifle

The Argent Adept:  Arcane Cadence, Arcane Cadence, Inspiring Supertonic, Alacritous Subdominant.

The Visionary:  Wrest the Mind, Twist the Ether, Precognition, Suggestion.

Nightmist:  Amulet of the Elder Gods, Mistform, Call Forth, anything with a {4}.

Darkwatch Nightmist:  Enlightenment, Master of Magic, Starshield Necklace, Tome of Elder Magic.

Bunker:  If I care only about winning, Flak Cannon, Grenade Launcher, Heavy Plating, Turret Mode.  If I don't want to be completely bored, these cards are at the bottom of my deck (except the Plating), and I do something like Omni-Cannon, Gatling Gun, Ammo Drop, and AuxPS, drawing DecommWare on my first Initialize.

Oh gawd, I get that all the gorram time.  No thanks!

Ra:  Imbued Fire, Flesh of the Sun God, Drawn to the Flame, Flame Blast

Dude, it's the best. You get 4 staves, PLUS an entire other 4-card hand to pick from. After that it doesn't really matter what other cards you get, they pretty much all do fire.

Fanatic:  Brutal Censure, Final Dive, Aegis of Resurrection, Wrathful Retribution

Redeemer Fanatic:  Absolution, Smite the Transgressor, Divine Focus, Prayer of Desperation.

Prime Wardens Fanatic (assuming she turns out as we've heard she will):  Sacrosanct Martyr, Embolden, Chastize, Holy Nova.

I hate the redundancy of drawing multiple Staffs and being forced to destroy them.  It's sometimes worth it for the healing or the extra 1 damage (or if you draw Flame Spike), but overall I'd rather do more interesting things.

well, you don't HAVE to destroy them. They're there for a buffer. In case the one gets destroyed or something. You always have extras to play.

Expat I'd go:  Shotgun, Shock Rounds, Speedloading, Assault Rifle.  Good against anyone.

DWExpat:  Hairtrigger, Shock Rounds, Assault Rifle, reload

Fanatic: (straight up murder) End of Days, Absolution, Embolden, Wrathful Retribution

Does she have three, or four? I started with three once - sadly the fourth card was an Inventory Barrage, not really of much use at the start of the game. But that was a pretty epic first turn, anyway :).

Grappling hook gives you destruction, and that has a lot of value.  I would take a third impromptu over micro though.

Omnitron-X:  Technological Advancement, Technological advancement, Sedative Flechettes, Bio Engineering beam.

The Scholar: (w/group heals) Keep Moving, Keep Moving, Keep Moving, Know when to hold fast.

The Scholar:  (w/o group heals) Keep Moving, Bring what you need, Transmutive Recovery, Proverbs and Axioms

She only has three, Tesla. Very few heroes have 4 copies of one card in their deck. The only one I can think of at the moment is Ra's Staff. A card as powerful as Impromptu isn't going to have more than 3 copies. Though if there 4 copies, it would be the best starting hand, since on top of putting equipment into play, you'd get several free draws and end up with one free play.

Omnix has 4x Reset, and if you think it can't be a powerful card, Argent Adept has 4x Arcane Cadence and Nightmist has 4x Oblivion.  Ab'Zero is probably the only hero who has two x4s, though (both his Modules).

Apart from Wrathful Retribution and the Adept's instruments, are there any hero cards of which there is only one copy? Not counting Nightmist's cards with their differing numbers as the text and card names are the same. I can't think of any, off the top of my head...

Chrono-Ranger's bounties and equipment are all unique, except for the Temporal Grenade.

Unity has Champion Bot, Cryo Bot, Stealth Bot, Swift Bot and Turret Bot.

There's some unique hero cards coming in Vengeance as well, but I can't say what or for whom.

In addition to the Arcane Cadences, The Argent Adept has 4 Instrumental Conjurations. As mentioned, AZ has four 4s; Chrono-Ranger can match this, with 4 each of Eye one the Prize, Just Doin' My Job, Sudden Contract and Displaced Armory, as well as his aforementioned unique bounties and equipment. Expat has Pride and Prejudice, which are unique. Wrathful Retribution is Fanatic's only 1, and she has no 4s. Fixer has 4 Overdrives and 4 Bloody Knuckleses. As mentioned before, Nightmist has 4 Oblivions. Omnitron-X has 4 Technological Advancements, as well as his 4 Resets. Ra has 4 Summon Staffs to go with his Staffs of Ra. The Scholar has 4 Keep Movings. Unity has 4 Brainstorms, as well as her five unique bots. The Visionary has 4 Mind Spikes. Wraith, Bunker, Legacy, Tachyon, Tempest and Haka have no 1s or 4s.

So the Freedom 4 have no 4s...nice.  Also, the two "honorary" Freedom Team members (as seen in the "team picnic" picture on Iron Legacy's one card) follow this pattern, but actual FV member AZ does not.  Quite thematic there.

Some of my personal maximum-fist-pump hands, if we were in a sitch where we chose the cards, and then randomly selected the villain/environment:

 

Legacy: Inspiring Presence, Lead From The Front, Next Evolution, Legacy Ring.

 

Argent Adept: That song that lets anyone play a card as a perform, that song that lets anyone but you play a card as an accompany, that instrument that lets you do both, and a Silver Shadow (I don't have AA's cards memorized but I know my favorite setup when I see it).

 

Scholar: 2xProverbs and Axioms, 2xDon't Dismiss Anything.

 

Chrono-Ranger: By Any Means, The Ultimate Target, Displaced Armory, Eye On The Prize

 

Tempest: Grevious Hailstorm, Cleansing Downpour, Gene-Bound Shackles, Into The Stratosphere

The "play a card" perform is Alacritous Subdominant, the "someone else play a card" accompany is Inventive Preparation. I think the Lyra is the instrument to play both of those.
