The Punishment ability from Relic allows you to treat a penalty as if it were a bonus when you attack the enemy that gave it to you. What happens if you have multiple penalties from multiple enemies and you attack all of them with the same die? Do they all turn into bonuses? Or is the effect die modified selectively for each target?
Also, when you use Frontline Fighting (Physical Powerhouse), does the target have to use the basic "Attack" action or any Attack action? I'd infer that "the Attack action" instead of "an Attack action" means basic attacks only, but if so, this would shut down villains hard.

For the multiple enemies + Punishment question, I would say that the die is modified selectively for each target.
For Frontline Fighting, since it does not specify a 'basic Attack action', I believe that the target can still use abilities. Their action(s) on their turn just need to include an Attack on/involving the user of Frontline Fighting

I would second catDreaming:
Punishment is probably only a bonus for that single enemy. Let's say you have a -1 penalty from Villain A, and you have a -2 penalty from Villain B, and there's also some minions. Then, you Attack multiple with the Mid die. You apply both penalties to the Mid die, so your Mid result is -3 against the minions, -1 against Villain A, and +1 Against Villain B. "Attack multiple" gets weird fast, but I think this is how I'd run it.
For Frontline Fighting, it says "the Attack action," and when any character performs an Action on their turn, that Action may be an Ability or Basic Action (by the steps outlined in Chapter 2). I assume enemies follow those exact steps, as well. So the Villain, if capable of the Attack action, must take it... but they can still use their Abilities, modifiers, etc. I don't see a distinction between "the Attack Action" and "an Attack Action" here, but I can totally see where you might infer it. I like strictly constructed language in gaming rules! I don't think these rules use very strictly constructed language, though, and I am okay with that. :)
